Addressing Challenges in Antibody Drug Conjugate Development
Antibody drug conjugates (ADCs) are a novel twist on conventional antibody therapeutics, driving more effective therapeutic responses with improved on-target specificities. ADCs typically consist of a monoclonal antibody attached to a cytotoxic payload via chemical linker. By conjugating a chemical payload onto an antibody, small highly toxic molecules can be precisely delivered to target tissue expressing a specific biomarker or cells presenting a certain target protein. Due to the wide variety of factors that influence the efficacy and safety of ADCs, optimizing steps starting from the selected biomarker target, monoclonal antibody quality, payload type, linker stability, and conjugation methods are necessary. In particular, achieving optimal drug to antibody ratios without compromising pharmacokinetics is a major obstacle towards ADC development. To overcome these challenges, ADC development is highly dependent upon interdisciplinary collaboration along with innovative technologies and solutions for success. Quality starts from the array of reagents, assays, and other solutions that are sourced. In this presentation, we present an overview of the entire ADC development workflow and highlight the challenges that need to be overcome alongside solutions designed to accelerate ADC development and overcome its major hurdles.
