The Hybridoma Screening Kit (His-Antigen) (TR-FRET) is based on a homogeneous (no wash) sandwich TR-FRET technology (Time-Resolved Fluorescence Resonance Energy Transfer) to screening for mouse hybridoma antibody binding to His-tagged antigen within 0.5-1 hours. It can also be used to rapidly identify the affinity of antibodies in the supernatant of mouse hybridoma cells.
Storage
1. Store the unopened kit at 2-8°C upon receipt.
2. Locate the expiration date on the outer packaging and do not use reagents beyond their expiration date.
3. The opened kit should be stored according to the storage conditions listed in the Components Table. The shelf life of the opened kit is 30 days from the date of opening.

This Hybridoma Screening Kit (His-Antigen) (TR-FRET) is based on TR-FRET technology (Time-Resolved Fluorescence Resonance Energy Transfer). A sandwich immunoassay was used to measure mouse hybridoma cell supernatant binding to a His-tagged antigen. This assay uses the Monoclonal Anti Mouse IgG-Europium chelate as the donor to bind to the mouse hybridoma cell supernatant, and FA Labeled Monoclonal Anti His Tag Antibody as the acceptor to bind to the His-tagged antigen.
— When the mouse hybridoma cell supernatant contains the target antibody, the donor and acceptor are in close proximity because of the binding of the mouse hybridoma antibody and its His-tagged target antigen. The 620 nm signal emitted by the donor under specific light source excitation is received by the acceptor, emitting a 665 nm signal.
— When the sample does not contain the target antibody, the donor and acceptor cannot get close to each other, thereby preventing FRET from occurring.

High-dose effect is a common phenomenon in TR-FRET assays. The optimal antigen concentration and antibody dilution must be determined empirically for each experimental setup.

The sample demonstrates a clear dose-dependent response across different matrices.

This kit is not only suitable for detecting Mouse IgG1, but also for Mouse IgG2a and Mouse IgG2b.
