Source

Rat Siglec-15 Protein, His Tag (SG5-R52H3) is expressed from human 293 cells (HEK293). It contains AA Arg 24 - Pro 257 (Accession # D3ZL05).

Predicted N-terminus: Arg 24

Molecular Characterization

This protein carries a polyhistidine tag at the C-terminus.

The protein has a calculated MW of 27.4 kDa. The protein migrates as 32-37 kDa when calibrated against Star Ribbon Pre-stained Protein Marker under reducing (R) condition (SDS-PAGE) due to glycosylation.

The protein is designed as a dimer.

Endotoxin

Less than 1.0 EU per μg by the LAL method / rFC method.

Purity

>90% as determined by SDS-PAGE.

Formulation

Lyophilized from 0.22 μm filtered solution in PBS, 0.5 M Arginine, pH7.4 with trehalose as protectant.

Background

Siglec-15 is a transmembrane glycoprotein belonging to the Siglec family, recognized for its sialic acid-binding properties and role in immune regulation. Structurally, its extracellular domain contains two Ig-like domains, and it functionally associates with the signaling adaptors DAP12 and DAP10. Siglec-15 is expressed on immune cells, including osteoclasts, macrophages, and dendritic cells. It exerts its function by binding to its ligand, the sialyl-Tn antigen. This interaction is critical for inducing TGF-beta production by tumor-associated macrophages and plays an essential role in osteoclast formation and TRANCE/RANK Ligand signaling.
