CAR Cell Activation and Expansion
The use of antigenic polymers based on magnetic bead coupled can effectively activate CAR-T cells with specificity.
CAR Cell In Vitro Analysis
After co-incubation with CAR-T cells using target-specific activation beads, in vitro analyses such as T cell activation, proliferation, and cytokine release were performed to locate product characterization and release testing.

Target-specific magnetic beads can be used to enrich low abundance CAR cells or rare CAR-T cells
BCMA CAR-T cells were stimulated with ActiveMax® Human BCMA μBeads (MBS-C004) for 24 h , and cell-free supernatants were harvested for evaluating IFN-γ、TNF-α、IL-4、IL-6 secretion by flow cytomtry. The results showed that BCMA CAR-T cells released significantly larger amounts of IFN-γ、TNF-α、IL-4、IL-6 into the supernatants in response to BCMA μBeads.

ActiveMax Streptavidin coupled μbeads, premium grade coupled with protein. 40ug of the biotinylated human BCMA protein (Cat. No. BCA-H82E4) was added to 1mg of ActiveMax Streptavidin coupled μbeads, premium grade (for cells) (Cat. No. MBS-C009), mix well and incubate for 60 mins at room temperature. And then the coupled μbeads were fluorescently stained using PE labeled anti-human BCMA antibody and analyzed by flow cytometry.

No. BCA-H82E4) in serial dose was added to 1mg of ActiveMax Streptavidin coupled μbeads, premium grade (for cells) (Cat. No. MBS-C009), mix well and incubate for 60 mins at room temperature. And then the unbound protein in supernatant measured by Micro BCA Protein Assay Kit was subtracted to calculate the BCMA protein coupled amount on the μbeads (loaded capacity).

And then the coupled μbeads were fluorescently stained using PE labeled anti-mouse Fc antibody and analyzed by flow cytometry.
