Thymic stromal lymphopoietin (TSLP) is an epithelial cell-derived cytokine involved in the pathology of inflammatory skin diseases, and is widely expressed by epithelial cells. Human TSLP cDNA encodes a 159 amino acid (aa) residue precursor protein with a 28 aa signal sequence (4, 5). Human TSLP has been shown to developing nondeletional central tolerance, amplifying epithelium-induced class switching, inducing atopic diseases and maintaining intestinal noninflammatory environment. Among diverse cells responding to Human TSLP, CD11c+ dendritic cells are the most obviously characterized target cells.

How should cross-species TSLP binding be evaluated for preclinical studies?

Compare human TSLP with mouse, cynomolgus, or rat TSLP according to the intended model. Use matched variants, tags, and assay conditions because sequence and construct differences can both affect measured affinity.

Why can SPR and BLI report different apparent affinities for TSLP–TSLPR binding?

Surface density, capture orientation, Fc-related avidity, and mass transport can shift the apparent affinity. Compare candidates within the same setup and use the second platform as orthogonal confirmation.

How can I distinguish a TSLP-binding antibody from a receptor-blocking antibody?

Confirm TSLP binding by ELISA, SPR, or BLI, then test inhibition of the TSLP–TSLPR–IL-7Rα interaction. Strong binding with weak inhibition suggests a non-blocking epitope or an unfavorable binding geometry.

Why should TSLP blockers be tested against receptor-complex assembly rather than TSLP–TSLPR binding alone?

TSLP first engages TSLPR and then recruits IL-7Rα to form the signaling complex. Pairwise binding identifies direct interaction, while a TSLP/TSLPR/IL-7Rα inhibition assay tests whether the candidate disrupts complex assembly.

When should wild-type TSLP or the R127A/R130A variant be used?

R127A/R130A removes a putative furin-cleavage site and can improve stability of the full-length protein. It is useful for reproducible binding assays, while wild-type TSLP is preferable when native sequence fidelity is the primary requirement.

We got the TSLP recombinant protein from your company. The plan was to stimulate cell in vitro to see if it can induce a TH2 response. Your company offers the most cost effective option for the TSLP protein and with good customer service and express shipping. So far we do see some level of responses in the cells, we will perform more testing to validate the effects. The quality of the protein seems quite good and worth trying.
