  • Evaluation of CAR expression

     B7-H3 (4Ig) CAR_T

    5e5 of anti-B7-H3 CAR-293 cells were stained with 100 μL of 1:50 dilution (2 μL stock solution in 100 μL FACS buffer) of PE-Labeled Human B7-H3 (4Ig) Protein, His Tag (Cat. No. B73-HP2H3) and negative control protein respectively(Fig. C and B), and non-transfected 293 cells were used as a control (Fig. A). PE signal was used to evaluate the binding activity (QC tested).

    5e5 of PBMCs were stained with PE-Labeled Human B7-H3 (4Ig) Protein, His Tag (Cat. No. B73-HP2H3) and anti-CD3 antibody, washed and then analyzed with FACS. FITC signal was used to evaluate the expression of CD3+ T cells in PBMCs , and PE signal was used to evaluate the non-specific binding activity to PBMCs (QC tested).

    What is the difference between Human B7-H3 and Human B7-H3 (4Ig)?

    The Human B7-H3 protein with an expression range of Leu29–Pro245 contains a single pair of immunoglobulin-like V and C domains. The product with an expression range of Gly27–Thr461, named B7-H3 (4Ig), includes the entire extracellular region.

Background

B7 homolog 3 (B7-H3), a member of the immunoglobulin superfamily, is also known CD276, which contains two Ig-like C2-type (immunoglobulin-like) domains and two Ig-like V-type (immunoglobulin-like) domains. B7-H3 may participate in the regulation of T-cell-mediated immune response. B7-H3 also plays a protective role in tumor cells by inhibiting natural-killer mediated cell lysis as well as a role of marker for detection of neuroblastoma cells. Furthermore, B7-H3 is involved in the development of acute and chronic transplant rejection and in the regulation of lymphocytic activity at mucosal surfaces. It could also play a key role in providing the placenta and fetus with a suitable immunological environment throughout pregnancy.
